Molecular Pathology: Cyclin dependent kinase 4 (CDK4) encodes a key regulator of the G1/S cell cycle checkpoint and is coamplified with MDM2 in over 90% of dedifferentiated liposarcomas. CDK4 binds with cyclin D to form active complexes, phosphorylates RB1 protein, prevents its interaction with E2F transcription factor and allows the cell to escape G1-S checkpoint, thereby promoting cell proliferation..
